The pattern is:
If the numbers of column are A,B,C,D
Column A,B,C,D

A^2-B^2=C
C^2-B^2=D
 
First column
 
3^2-2^2=9-4=5
5^2-2^2=25-4=21
 
Second column
 
5^2-4^2=25-16=9
9^2-4^2=81-16=65

Third column
3^2-1^2=9-1=8
Missing number is 8

Fourth column

7^2-3^2=49-9=40
Missing number is 40
